Reading age from your lipids

Max Unfried is a longevity researcher who works on one of the most important, least glamorous problems in the whole field: how do you actually measure how old someone is on the inside? Chronological age is easy; biological age is the number that matters — and you can only improve what you can measure.

We first met Max very early on the quest, at the Longevity Summit in Dublin in 2023, while he was completing his doctoral research in Brian Kennedy’s lab in Singapore. His particular angle is unusual and, he argues, badly overlooked: lipids.

Most people hear “lipids” and think fats. But the class is much larger — steroids, cannabinoids and more — and lipids do an enormous amount of work in the body: signalling, energy storage, and the structural stability of every cell membrane. As Max points out, your brain is mostly lipids — and yet, as a window into ageing, they are barely studied compared with the DNA-based epigenetic clocks that get all the attention.

His work is to change that: to build lipid-based measures of biological age as a new modality — sitting alongside epigenetic age, proteomic age and the frailty indexes — and then to use them not just to read ageing but to modulate it, pointing toward new therapies. It is a quietly foundational contribution: better clocks are what let everyone else prove whether an intervention actually works.

He opened with something that stuck with us: that travelling the world with your kid, learning things that help you live longer, is itself a longevity intervention. Why biological age

Chronological age tells you how many birthdays you have had; biological age tells you how your body is actually doing. It is the number longevity science is really chasing — because you can only improve what you can measure, and every claimed intervention has to be judged against a good clock.

The bet on lipids

Most ageing clocks read DNA methylation. Max works one layer over: lipids — the fats, steroids and signalling molecules that make up cell membranes and most of your brain. They are central to how cells work and, he argues, badly under-used as a marker of ageing. Lipid-based clocks could be a whole new modality.

On travelling with your son

Max Unfried“Fantastic that you guys travel together. I think more parents should do that with their kids. School is important — but taking a year off to learn things for life, things that might help you live longer anyway, can be very important too.”

Max Unfried“What I am excited about is using those lipids to measure biological age — as a new modality, in addition to epigenetic age, proteomic age and the frailty indexes — so we understand what happens with lipids in ageing, and then modulate them: to find new therapies that target lipids, and new lipids that can be used to help.”
